Prin ATP glicerolul si glicerolul kinaza comduc la compusii chimici ADP si glicerol 3 fosfat. Glicerol 3-fosfatul este oxidat prin glicerol kinaza si se obtin dihidroxiacetona fosfat si peroxid hidrogen. Peroxid hidrogenul combinat cu fenol si 4-aminoantipirin conduce la un complex chimic colorat. Intensitatea culorii masurata fotometric este direct proportionala cu concentratia de trigliceride.CARACTERISTICILimita de detectie: 2 mg/dl (0,02 mmol/l)Sensitivitate: 0,65 mA x dl / mg (57 mA x l / mmol)Liniaritate: 1000 mg/dl (11,3 mmol/l).
